| Abstract: |
| The aim is to enhance the reusability of lipase, thereby promoting the application of functional lipids.The lipase was immobilized on macroporous resin, and the immobilization conditions were optimized through single-factor experiments and response surface methodology. The prepared immobilized lipase was applied to the preparation of avocado diacylglycerol oil, and the fatty acid composition of avocado diacylglycerol oil was analyzed, and its reusability and storage stability were analyzed. The results showed that the optimal immobilization conditions for the lipase were macroporous resin D101 as the carrier, lipase mass concentration 0.21 g/mL (10 g carrier), immobilization time 5 h, immobilization temperature 41 ℃, and phosphate buffer solution pH 7.5. Under these conditions, the enzyme activity of the immobilized lipase was 6.871 U/mg. The immobilized lipase was used to prepare avocado diacylglycerol oil, and the content of diacylglycerol reached 81.42%. Moreover, the fatty acid composition of avocado diacylglycerol oil and avocado oil were consistent. The enzyme activity remained at 62.2% of the initial activity after 6 times consecutive uses, and 92.1% of the initial activity after 30 d of storage at 4 ℃. In conclusion, the immobilized lipase prepared by adsorption can be applied in the production of avocado diacylglycerol oil without altering the original fatty acid composition of avocado oil. |
