      运用超声辅助90%乙醇溶液提取芝麻素。在单因素实验的基础上,运用正交实验优化芝麻素超声辅助提取工艺参数。普鲁士蓝法测定芝麻素还原能力,滤纸片-抑菌圈法测定其抗菌活性。结果表明:超声辅助提取的最优条件为超声功率300 W、液固比25∶1、超声温度55℃、超声时间25 min,在最优条件下,芝麻素提取率为(3.942±0.085)mg/g;芝麻素具有一定的还原能力,但比相同质量浓度的VC弱得多;当质量浓度大于40 mg/mL时,芝麻素对蜡状芽孢杆菌、金黄色葡萄球菌、枯草芽孢杆菌、大肠杆菌均有不同程度的抑制作用,质量浓度大于60 mg/mL时其对4种细菌抑制作用的强弱顺序为蜡状芽孢杆菌>金黄色葡萄球菌>枯草芽孢杆菌>大肠杆菌,而当质量浓度小于100 mg/mL时,芝麻素对真菌黄曲霉无抑制作用。

      Sesamin was extracted by ultrasound-assisted 90% ethanol solution. On the basis of single factor experiment, the parameters of the ultrasound-assisted extraction of sesamin were optimized by orthogonal experiment. The reducing activity and the antibacterial activity of sesamin were determined by Prussian blue method and filter paper-inhibition zone method, respectively. The results showed that the optimal ultrasound-assisted extraction conditions were obtained as follows:ultrasonic power 300 W, ratio of liquid to solid 25∶1, ultrasonic temperature 55℃ and ultrasonic time 25 min. Under the optimal conditions, the extraction rate of sesamin was (3.942±0.085) mg/g. Sesamin had certain reducing activity, much weaker than VC of the same mass concentration. When the mass concentration of sesamin was higher than 40 mg/mL, sesamin showed different degrees of inhibitory action on Bacillus cereus,Staphylococcus aureus,Bacillus subtilis and Escherichia coli, and the inhibitory action of sesamin with mass concentration higher than 60 mg/mL on Bacillus cereus was the strongest, followed by Staphylococcus aureus,Bacillus subtilis and Escherichia coli. When the mass concentration of sesamin was less than 100 mg/mL, no obvious inhibitory action of sesamin on Aspergillus flavus was observed.
