为进一步提高终极腐霉EPA产量,在初步优化发酵条件的基础上,通过Plackett-Burman和Box-Behnken实验设计优化终极腐霉生产EPA发酵工艺。获得的最优发酵工艺条件为:蔗糖8%,硝酸钾0.4%,酵母粉0.65%,磷酸氢二钠0.175%,硫酸镁0.065%,大豆油1.0%,起始pH 6.0,装液量50 mL/250 mL。在最优发酵工艺条件下,EPA产量达到541.61 mg/L,比初步优化产量(456.39 mg/L)提高了85.22 mg/L。 |

In order to increase the EPA yield produced by Pythium ultimum, the fermentation process of Pythium ultimum for EPA production was optimized by Plackett-Burman design combined with Box-Behnken design on the basis of preliminary optimal fermentation conditions. The results showed that the optimal fermentation conditions were obtained as follows: sucrose 8%, KNO3 0.4%, yeast powder 0.65%, Na2HPO40.175%, MgSO40.065%, soybean oil 1.0%, initial pH 6.0, liquid volume in flask 50 mL/250 mL.Under the optimal conditions, EPA yield reached 541.61 mg/L, which increased by 85.22 mg/L compared with that of preliminary optimization (456.39 mg/L). |
