      研究栀子油对四氯化碳所致小鼠急性肝损伤的保护作用,并探讨其保肝作用机制。采用四氯化碳小鼠急性肝损伤模型,测定小鼠血清中ALT、AST活性,TBA、TBIL、DBIL水平,肝组织中SOD活力及MDA、GSH的水平,用HE染色切片对肝组织进行病理学观察,考察栀子油各剂量组对四氯化碳所致小鼠急性肝损伤的保护作用。结果表明:与模型组相比,栀子油中剂量组(0.6 g/kg)、高剂量组(1.2 g/kg)可显著改善四氯化碳所致急性肝损伤小鼠的肝脏组织病理学变化,显著降低肝损伤小鼠血清中ALT、AST的活性和TBA、TBIL、DBIL的水平;栀子油高剂量组可提高肝匀浆中SOD活力和GSH水平,降低MDA含量。栀子油对四氯化碳所致急性肝损伤小鼠具有保肝作用,其机制可能与降酶、抑制自由基脂质过氧化、维持肝细胞膜正常形态有关。

      The protective effect of Gardenia oil on carbon tetrachloride acute liver injury in mice was studied and its protective mechanism was investigated. The activities of ALT, AST and levels of TBA, TBIL, DBIL in serum, the activities of SOD and levels of MDA and GSH in liver tissue were measured and pathological morphological in liver tissues were monitored by HE staining section in carbon tetrachloride acute liver injury mice. The protective effect of different doses group of Gardenia oil on carbon tetrachloride acute liver injury in mice was discussed. The results showed that compared with model group, middle dose group (0.6 g/kg) and high dose group (1.2 g/kg) of Gardenia oil could significantly improve the pathological changes of liver tissue in carban tertrachloride acute liver injury mice, and could significantly reduce the activities of ALT, AST and the levels of TBA, TBIL, DBIL in serum. High dose group (1.2 g/kg) of Gardenia oil could improve the SOD activity and GSH level, and reduce the MDA content in liver homogenate. Gardenia oil had a significant hepatoprotective effect on carbon tetrachloride induced liver injury mice. The mechanism might be related to enzyme decreasing, inhibition of free radical lipid peroxidation and maintenance of normal morphology of hepatic cells.
