      建立了反相高效液相色谱-蒸发光散射(HPLC-ELSD)法测定肌醇脂肪酸酯制备过程中肌醇含量。利用水对样品进行萃取定容,采用Agilent ZORBAX 300SB-C18 (4.6 mm×250 mm,5 μm) 色谱柱,甲醇和0.1%冰乙酸溶液作为流动相,在0.8 mL/min流速下进行梯度洗脱,蒸发器温度55 ℃,N2压力206.84 kPa,增益10,以外标法对肌醇进行定量。结果表明:该方法在0.01~1.00 mg/mL质量浓度范围内线性良好,标准曲线回归方程相关系数(R2)为0.993 4,加标回收率为80%~108%,相对标准偏差均小于6%。该方法前处理简单、重复性好、灵敏度高、结果准确可靠,可应用于肌醇脂肪酸酯制备过程中肌醇的检测。

      A reverse-phase high performance liquid chromatography with evaporative light scattering detector (HPLC-ELSD) method was developed for the analysis of inositol during the preparation of inositol fatty acid esters. The sample was extracted and diluted with water for subsequent analysis. The Agilent ZORBAX 300SB-C18 (4.6 mm×250 mm,5 μm) was utilized. The mobile phase was methanol solution containing 0.1% acetic acid and the flow rate was 0.8 mL/min. Evaporator temperature was 55 ℃ and N2 pressure was 206.84 kPa with the gain 10. Inositol was quantified by external standard method. The results showed that the method had good linearity for inositol in the range of 0.01-1.00 mg/mL and R2 was 0.993 4. The recoveries of inositol were in the range of 80%-108% with the relative standard deviation less than 6%. This method was simple, stable, sensitive and reliable. It could be applied to the determination of inositol during the preparation of inositol fatty acid esters.
