为明确奇数链脂肪酸(OCFA)与人体健康的关系,采用气相色谱-质谱法(GC-MS)检测分析健康人群和结肠癌患者血清中OCFA的分布特征差异,并采用脂多糖(LPS)诱导的Caco-2细胞炎症模型探究OCFA对炎症的抑制效应,包括对细胞活性、促炎因子基因表达以及脂肪酸组成的影响。结果表明:健康人群中的OCFA(C15∶ 0和C17∶ 0)含量显著高于结肠癌人群;男性以及高龄(59岁以上)结肠癌人群中C15∶ 0和C17∶ 0水平偏低;C15∶ 0和C17∶ 0对细胞活力无影响,C15∶ 0能够显著降低LPS刺激后Caco-2细胞中促炎因子TNF-α、IL-8和iNOS的表达,C17∶ 0能够显著降低促炎因子IL-6、TNF-α、IL-8和iNOS的表达,两种OCFA均能够起到类似于n-3 PUFA缓解炎症刺激的作用;两种OCFA(C15∶ 0和C17∶ 0)能够在肠道细胞Caco-2中吸收并代谢成C17∶ 0、C17∶ 1。综上,OCFA能够调节肠道炎症因子的表达进而抑制肠上皮细胞炎症反应,对肠道健康有潜在的保护作用。 |

In order to explore the health effects of odd-chain fatty acids (OCFA), serum OCFA levels in the healthy individuals and colon cancer patients were measured by gas chromatography-mass spectrometry (GC-MS) to investigate the differences in the distribution characteristics, and the protective effects of OCFA on inflammation by using the lipopolysaccharide (LPS)-induced Caco-2 cell inflammation model were studied, including the changes in cell activity, pro-inflammatory factor gene expression and fatty acid composition. The results showed that the levels of two main OCFA (C15∶ 0 and C17∶ 0) in healthy individuals were significantly higher than that in colon cancer patients.The levels of C15∶ 0 and C17∶ 0 in male patients and patients over the age of 59 were significantly lower. C15∶ 0 and C17∶ 0 had no effect on cell activity. C15∶ 0 could significantly inhibit the expression of pro-inflammatory factors TNF-α, IL-8 and iNOS in Caco-2 cells with LPS stimulation, C17∶ 0 could significantly reduce the expression of IL-6, TNF-α, IL-8 and iNOS. Two main OCFA could play a similar role in alleviating inflammatory stimulation as n-3 PUFA. Two main OCFA could be easily incorporated by Caco-2 cells into C17∶ 0 and C17∶ 1 in Caco-2. In conclusion, OCFA may have a potential preventive effect on inflammation and intestinal health by regulating the expression of pro-inflammatory cytokines. |
