      旨在为葡萄球菌JJU-412的工业化产胞外脂肪酶、餐厨油污治理和资源化利用,及其胞外脂肪酶的应用提供一定的理论基础和技术支持,对JJU-412产胞外脂肪酶的非灭菌发酵条件进行单因素实验优化,并与灭菌发酵进行比较,在此基础上进一步探究其煎炸废油降解性能和表征其酶学性质。结果表明:JJU-412的最优非灭菌发酵产胞外脂肪酶条件为发酵培养基pH 9、发酵温度25 ℃、装样量30 mL/250 mL、接种量3%(以发酵培养基体积计)和发酵时间20 h,在此条件下非灭菌发酵产酶活力与灭菌发酵相比没有显著差异;在最优非灭菌发酵培养基pH、发酵温度、装样量及接种量下发酵28 h,JJU-412产胞外脂肪酶对煎炸废油的降解率达到85.79%;JJU-412产胞外脂肪酶粗酶可特异性水解短中链对硝基苯酚酯,最适反应温度和pH分别为40 ℃和9,为中温碱性非金属依赖性脂肪酶,具有一定的温度稳定性和良好的pH稳定性,具有良好的有机溶剂和表面活性剂耐受性。综上,JJU-412可通过非灭菌发酵产胞外脂肪酶,并可高效降解煎炸废油,其胞外脂肪酶具有优良的酶学性质。

      To provide a certain theoretical basis and technical support for the industrial production of extracellular lipase by Staphylococcus sp. JJU-412, as well as its potential application in the treatment and resource utilization of kitchen oil pollution and the application of its extracellular lipase, the non-sterile fermentation conditions of extracellular lipase produced by JJU-412 were optimized through single-factor experiments,and compared with sterile fermentation. Based on this, its degradation performance in waste frying oil was investigated,and its enzymatic properties were characterized. The results indicated that the optimal non-sterile fermentation conditions for extracellular lipase produced by JJU-412 were obtained as follows: fermentation medium pH 9, fermentation temperature 25 ℃, sample loading amount 30 mL/250 mL, inoculum size 3%(based on the volume of fermentation medium), and fermentation time 20 h.Under these conditions, there was no significant difference in enzyme activity between non-sterile and sterile fermentation. When fermented for 28 h under the optimal non-sterile fermentation conditions of fermentation medium pH, fermentation temperature, sample loading amount, and inoculum size, the degradation rate of waste frying oil by the extracellular lipase produced by JJU-412 reached 85.79%. The crude extracellular lipase produced by JJU-412 specifically hydrolyzed short- to medium-chain p-nitrophenol esters, with optimal reaction temperature 40 ℃ and pH 9, respectively. It was a medium temperature, alkaline, and non-metal-dependent lipase that exhibited moderate thermal stability and excellent pH stability, and good tolerance to organic solvents and surfactants. In summary, JJU-412 can produce extracellular lipase through non-sterile fermentation and efficiently degrade waste frying oil, as well as its extracellular lipase exhibits excellent enzymatic properties.
