| 旨在为亚麻籽油的“减苦增稳”策略提供依据,以低温压榨亚麻籽油为研究对象,通过添加混合环肽和γ-生育酚,建立不同的Schaal烘箱加速氧化模型,探究加速氧化过程中其氧化指标和环肽O、环肽D、γ-生育酚含量的变化。同时,对环肽O单体进行体外Rancimat加速氧化实验,并结合液相色谱-质谱/质谱(LC-MS/MS)分析,考察γ-生育酚对含有色氨酸残基的环肽O氧化产物的影响。结果表明:当环肽O上蛋氨酸(Met)残基尚未氧化时,γ-生育酚对其起抗氧化作用;当环肽O上的Met被氧化为蛋氨酸亚砜(MetO)即环肽D后,γ-生育酚对环肽D起促氧化作用;长期氧化后的环肽D仅有痕量的蛋氨酸砜(MetO2)形式,大部分裂解为四肽和五肽的形式,且γ-生育酚促进了环肽D的碎裂。综上,γ-生育酚在亚麻籽油氧化过程中对环肽O具有抗氧化作用,但当环肽O的Met氧化为MetO形成环肽D后,其作用转变为促氧化,并促进环肽D的裂解。 |

| To provide a basis for the "bitterness reduction and stability enhancement" strategy in flaxseed oil, the cold-pressed flaxseed oil was used as the research subject, cyclolinopeptide mixture and γ-tocopherol were added into flaxseed oil, and different Schaal oven accelerated oxidation models were established to explore the changes of oxidative indexes as well as the contents of cyclolinopeptide O(CLO), cyclolinopeptide D(CLD), and γ-tocopherol during accelerated oxidation. Additionally, an in vitro Rancimat accelerated oxidation experiment was conducted on CLO monomer, combined with LC-MS/MS analysis, to examine the effect of γ-tocopherol on the oxidation products of CLO with tryptophan residue. The results indicated that γ-tocopherol exerted an antioxidant effect on CLO when its methionine (Met) residue was unoxidized. However, once Met residue in CLO was oxidized to methionine sulfoxide (MetO, CLD), γ-tocopherol promoted the oxidation of CLD. After prolonged oxidation, CLD was mostly fragmented into tetrapeptides and pentapeptides, with only trace amounts of methionine sulfone(MetO2) forms remaining, and γ-tocopherol was found to facilitate the fragmentation of CLD. In conclusion, γ-tocopherol exhibits an antioxidant effect on CLO during the oxidation of flaxseed oil, but its role shifts to pro-oxidant upon oxidation of Met residue to MetO forming CLD, concurrently promoting the fragmentation of CLD. |
