Abstract:
      In order to develop hemp seed protein for high-value utilization, hemp seed alcohol dehydrogenase(ADH)-activating peptides were prepared by enzymolysis of hemp seed protein with protease.The ADH activation rate was used as the evaluation index to screen the best protease. On this basis, single-factor experiments and response surface methodology were adopted to optimize the preparation process of hemp seed ADH-activating peptides. The molecular weight distribution and amino acid composition of hemp seed ADH-activating peptides were determined, and the microstructure was analyzed by scanning electron microscope. The results showed that hemp seed ADH-activating peptides obtained by enzymolysis with neutral protease had the highest ADH activation rate and were more suitable for the enzymolysis of hemp seed protein. The optimal preparation conditions of hemp seed ADH-activating peptides were as follows: enzymolysis temperature 44 ℃, enzymolysis time 2.75 h, pH 62, enzyme dosage 3.36%. Under these conditions, the ADH activation rate was 63.14%. Hemp seed ADH-activating peptides were mainly small peptides with molecular weight less than 3 000 Da, accounting for 84.66%. Hemp seed ADH-activating peptides had a rich amino acid composition, and glutamic acid content was the highest (20.37%), followed by aspartic acid (1185%) and arginine(1170%). Hemp seed ADH-activating peptides had irregular shape, smooth surface and multi-pore structure. In conclusion, hemp seed ADH-activating peptides have a relatively high ADH activation rate and great potential in the development of alcohol-relieving and liver-protecting peptide products.
