| 旨在为工业化放大生产豆甾醇单体提供参考,以大豆油脱臭馏出物中提取的植物甾醇为原料,采用溶剂结晶法对其中的豆甾醇单体进行分离纯化,并通过单因素实验对豆甾醇分离纯化工艺条件进行优化。结果表明:豆甾醇单体的最优制备工艺条件为以正丁醇为溶剂,养晶温度8 ℃,养晶时间12 h,第1~6次重结晶的料溶比分别为1∶ 2、1∶ 3、1∶ 4、1∶ 4、1∶ 5、1∶ 6,第7次采用无水乙醇进行重结晶以消除正丁醇溶剂的影响,第7次料溶比为1∶ 10。在最优工艺条件下,豆甾醇含量由原料的29.15%提升至产品的96.04%,豆甾醇回收率为5.99%。综上,采用溶剂重结晶法可以获得高纯度的豆甾醇单体产品。 |

| To provide a reference for the industrial-scale production of stigmasterol monomer, phytosterols extracted from soybean oil deodorizer distillate were used as raw material, the stigmasterol monomer was purified by solvent crystallization, and the purification process conditions were optimized through single-factor experiments. The results showed that the optimal preparation conditions for stigmasterol monomer were as follows: using n-butanol as the solvent, crystallization temperature 8 ℃, crystallization time 12 h, and material-solvent ratios for the 1st to 6th recrystallization steps being 1∶ 2, 1∶ 3, 1∶ 4, 1∶ 4, 1∶ 5, and 1∶ 6, respectively. Anhydrous ethanol was used for the 7th recrystallization to eliminate the influence of n-butanol solvent, with a material-solvent ratio of 1∶ 10. Under these optimal conditions, the stigmasterol content increased from 29.15% in the raw material to 96.04% in the product, with a stigmasterol recovery rate of 5.99%. In conclusion, high-purity stigmasterol monomer product can be obtained using the solvent recrystallization method. |
