      旨在完善樟树籽仁油的食用安全性评价,研究了樟树籽仁油对受孕Wistar大鼠的毒性和胎鼠致畸性。根据GB 15193.14—2015《食品安全国家标准 致畸试验》,以大豆油为阴性对照,0.93、1.85、3.70 g/kg樟树籽仁油为低、中、高剂量组,于孕鼠妊娠期6~15 d连续灌胃,观察母体体征和体质量变化,解剖观察孕鼠胚胎形成指标和胎鼠生长发育指标,分析樟树籽仁油对胎鼠外观、骨骼发育和内脏发育的影响。结果表明:樟树籽仁油各剂量组孕鼠一般状况正常,未出现中毒特征;与阴性对照组相比,各剂量组孕鼠的体质量、体质量增量、体质量净增量、黄体数、着床数、活胎数、吸收胎数、死胎数均无显著性差异(p>0.05);各剂量组胎鼠的体质量、体长、外观畸形率、骨骼畸形率和内脏畸形率也没有显著性差异(p>0.05)。综上,樟树籽仁油未观察到有害作用剂量(NOAEL)为3.70 g/kg,对Wistar大鼠未见明显母体毒性、胚胎毒性和致畸性。

      To improve the safety assessment of Cinnamomum camphora seed kernel oil (CCSKO) for human consumption, its toxicity on Wistar pregnant rats and teratogenicity in fetal rats was studied. According to the Teratogenicity Test in Chinese National Standard for Food Safety (GB 15193.14-2015), with soybean oil as negative control, 0.93, 1.85 g/kg and 3.70 g/kg of CCSKO were used as the low-, middle- and high-dose groups to administrate via continuous gavage pregnant rats from gestation 6 d to 15 d, and the pregnant rats′ embryo signs and body weight changes and fetal rats′ growth and development indexes were observed by anatomy, and the effects of CCSKO on the appearance, skeletal development and internal organ development of fetal rats were analyzed. The results showed that pregnant rats in each dose group were normal, and without signs of toxicity. Compared with the negative control group, the body weight, weight gain, net weight gain, corpus luteum number, implantation number, live fetus number, absorbed fetus number and stillbirths′ number of pregnant rats in each dose group had no significant difference (p>0.05). There was no significant difference in the weight, body length, appearance deformity rate, skeletal development deformity rate and internal organ development deformity rate of fetal rats in each dose group (p>0.05). In conclusion, no-observed-adverse-effect level (NOAEL) of CCSKO is observed at 3.70 g/kg, and there is no obvious maternal toxicity, embryotoxicity and teratogenicity to Wistar rats.
