| 为了探究磷脂酶D(PLD)催化合成二硬脂酰磷脂酰乙醇胺(DSPE)的可行性,采用PLD催化二硬脂酰磷脂酰胆碱(DSPC)合成DSPE,并通过乙酸乙酯洗涤和柱层析法进行纯化,通过单因素实验对工艺条件(有机溶剂种类、pH、反应温度、有机相与水相体积比、酶用量、底物物质的量比、反应时间)进行优化。结果表明:酶法合成DSPE的最佳反应条件为以二氯甲烷为有机溶剂、pH 5.5、反应温度30 ℃、有机相与水相体积比1∶ 1、酶用量60%(以DSPC质量计)、底物乙醇胺与DSPC物质的量比40∶ 1、反应时间12 h,在此条件下DSPE的产率为92.1%;DSPE成品的纯度为9922%。综上,该工艺条件温和,产率高,可用于DSPE的制备。 |

| In order to explore the feasibility of phospholipase D (PLD) catalyzed synthesis of distearoylphosphatidylethanolamine (DSPE), the DSPE was synthesized from distearoylphosphatidylcholine (DSPC) catalyzed by PLD, and the resulting product was purified via ethyl acetate washing followed by column chromatography. The process conditions (organic solvent, pH, reaction temperature, volume ratio of organic phase to aqueous phase, enzyme dosage, substrate molar ratio, and reaction time) were optimized through a single-factor experiment. The results showed that the optimum reaction conditions for enzymatic synthesis of DSPE were as follows: using dichloromethane as the organic solvent, pH 5.5, reaction temperature 30 ℃, volume ratio of organic phase to aqueous phase 1∶ 1, enzyme dosage 60% (based on the mass of DSPC), molar ratio of ethanolamine to DSPC 40∶ 1, and reaction time 12 h. Under the optimum reaction conditions, the yield of DSPE was 92.1% and the purity of the finished product DSPE was 99.22%. In summary, this process offers mild conditions and high yields, making it suitable for the preparation of DSPE. |
