      为了对水酶法提取酸枣仁油的工业化应用提供参考,以酸枣仁为原料,通过单因素试验和响应面试验对水酶法提取酸枣仁油的工艺条件进行优化,并将水酶法提取的酸枣仁油与索氏提取法和超声辅助提取法提取的酸枣仁油的体外抗氧化活性和脂肪酸组成进行比较。结果表明:水酶法提取酸枣仁油的最优工艺条件为料液比1∶ 2、碱性蛋白酶添加量3%、酶解温度40 ℃、酶解时间9 h、酶解 pH 10,在此条件下酸枣仁油提取率达54.84%;与其他2种工艺提取的酸枣仁油相比,水酶法提取的酸枣仁油体外抗氧化活性最强,其对DPPH自由基清除率的IC50值为7.27 mg/mL,对ABTS+自由基清除率的IC50值为7.17 mg/mL;3种工艺提取的酸枣仁油的脂肪酸组成存在显著差异,但不饱和脂肪酸和饱和脂肪酸含量无显著差异。综上,水酶法可以高效提取酸枣仁油,且提取的酸枣仁油表现出较强的体外抗氧化活性。

      To provide a reference for the industrial application of aqueous enzymatic extraction(AEE) of wild jujube seed oil, the wild jujube seed was used as material, and the AEE conditions were optimized through single-factor experiments and response surface methodology(RSM). A comparative analysis of the in vitro antioxidant activity and fatty acid composition of the oils extracted by AEE, Soxhlet extraction(SE), and ultrasound-assisted extraction(UAE) was conducted. The results showed that the optimal conditions for AEE were as follows: solid-liquid ratio 1∶ 2, alkaline protease dosage 3%, enzymatic hydrolysis temperature 40 ℃, enzymatic hydrolysis time 9 h, and enzymatic hydrolysis pH 10. Under these conditions, the extraction yield of wild jujube seed oil reached 54.84%. Compared to the oils extracted by the other two methods, the oil obtained by AEE exhibited the strongest in vitro antioxidant activity, with IC50 values of 7.27 mg/mL for DPPH radical scavenging and 7.17 mg/mL for ABTS+ radical scavenging. Although the fatty acid composition of the oils extracted by the three methods showed significant differences, the contents of unsaturated and saturated fatty acids did not differ significantly. In conclusion, AEE is an efficient method for obtaining wild jujube seed oil, and the extracted oil demonstrates strong in vitro antioxidant activity.
