      为提高没食子酸的脂溶性,促进其作为抗氧化剂在食用油中的应用,在无溶剂体系中,以椰子油与没食子酸丙酯(PG)为反应底物,通过Lipozyme 435脂肪酶催化合成脂肪酰基没食子酸甘油酯(FAGG)。通过单因素实验研究了底物比(PG与椰子油物质的量比)、酶添加量、反应温度和反应时间对FAGG合成的影响,确定了最适反应条件,并比较了不同植物油(椰子油、亚麻籽油、高油酸葵花籽油)与PG在最适条件下的反应规律。结果表明:PG与椰子油合成FAGG的最适反应条件为底物比1∶ 25、酶添加量25%(以底物总质量计)、反应温度80 ℃、反应时间120 h,在此条件下FAGG产率为87.35%;3种植物油为底物时,FAGG的产率大小排序为椰子油>亚麻籽油>高油酸葵花籽油,表现为中链脂肪酸为主的植物油反应活性高于不饱和程度高的长链脂肪酸为主的植物油,并且在长链甘油三酯主要链长相同时,双键数目越多,反应活性越强。综上,中链脂肪酸油脂是酶促合成没食子酸结构脂中较为适宜的反应受体。

      To improve the lipophilicity of gallic acid and promote its application as an antioxidant in edible oils, a solvent-free system was employed in which coconut oil and propyl gallate (PG) served as substrates for the lipase-catalyzed synthesis of fatty acyl galloylglycerols (FAGG) using Lipozyme 435. Single-factor experiments were conducted to investigate the effects of substrate molar ratio (ratio of PG to coconut oil), enzyme loading, reaction temperature, and reaction time on FAGG synthesis, and the optimal reaction conditions were determined. In addition, the reaction behaviors of different vegetable oils (coconut oil, flaxseed oil, and high-oleic acid sunflower seed oil) with PG were compared under these optimal conditions. The results showed that the optimal conditions for synthesizing FAGG from PG and coconut oil were a substrate molar ratio of 1∶ 25, enzyme loading of 25% (based on the total mass of the substrate), reaction temperature of 80 ℃, and reaction time of 120 h, under which the FAGG yield reached 87.35%. When three different vegetable oils were used as substrates, the FAGG yield followed the order: coconut oil>flaxseed oil>high-oleic acid sunflower seed oil, which indicated that oils rich in medium-chain fatty acids exhibited higher reactivity than that rich in long-chain fatty acids with a high degree of unsaturation. Furthermore, for triacylglycerols with similar long-chain lengths, a higher number of double bonds corresponded to greater reaction activity. In summary, medium-chain fatty acid-rich oils are more suitable acyl acceptors for the enzymatic synthesis of gallic acid-based structured lipids.
