      为了提高热带海洋富油微藻Desmodesmus sp.WC08的油脂产率,采用响应面法对其自养培养条件进行优化研究。通过单因素试验初步确定了影响Desmodesmus sp.WC08油脂产率的9个因素的最佳条件。利用Plackett-Burman试验从中筛选出了显著影响Desmodesmus sp.WC08油脂产率的4个因素,运用Box-Behnken试验原理及响应面分析确定了4个因素的最佳参数为:初始OD680 0.33,氮浓度10 mmol/L,盐度1.6%和初始pH 6.5。在优化后的条件下培养Desmodesmus sp.WC08的油脂产率为141.47 mg/(L·d),较优化前提高了147.33%。
英文摘要:
      In order to improve the tropical rich-in-oil microalgae Desmodesmus sp. WC08 oil yield, the response surface methodology was adopted for autotrophic study on optimization of culture conditions. By single factor experiment, nine factors optimal concentrations which could promote oil yield of Desmodesmus sp. WC08 were obtained. Based on Plackett-Burman experiment, four significant factors were screened owing to Desmodesmus sp. WC08 oil yield. The optimal conditions were determined through the Box-Behnken experiment and response surface methodology as follows: nitrogen concentration 10 mmol/L, salinity 1.6%, initial OD680 0.33, initial pH 6.5. Under the optimal conditions for cultivating Desmodesmus sp. WC08, the oil yield was 141.47 mg/(L·d), increased by 147.33% than that without optimization.
