      以单针藻Monoraphidium sp. QLY-1为研究对象,研究了外源褪黑素(MT)对缺氮条件下单针藻生物量、油脂含量、碳水化合物及蛋白质含量、活性氧(ROS)和丙二醛(MDA)含量以及3种主要抗氧化酶(超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT))活性的影响。结果表明:在外源MT的处理下,藻细胞中油脂含量是对照组的1.22倍,最高达到51.38%;蛋白质和碳水化合物含量分别从20.82%和37.69%下降到14.73%和 25.24%;此外,MT的添加降低了藻细胞内ROS和MDA的含量,上调了3种抗氧化酶活性,从而缓解了藻细胞在缺氮条件下导致的氧化性损伤。说明外源MT在缺氮条件下进一步促进单针藻中油脂的积累与调控ROS含量以及抗氧化酶活性有关,为提高微藻中油脂的合成提供了一种新的策略。

      With the microalga Monoraphidium sp. QLY-1 as the research object, the effects of exogenous melatonin (MT) on the biomass, lipid content, contents of carbohydrate and protein, reactive oxygen species (ROS), malondialdehyde (MDA) and activities of three key antioxidant enzymes, including superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) in Monoraphidium sp. QLY-1 under nitrogen deficiency condition were investigated. The results showed that affter the treatment of exogenous MT, the lipid content in algal cells was 1.22 times as high as that of the control, and the highest lipid content reached 51.38%. The contents of protein and carbohydrate decreased from 20.82% and 37.69% to 14.73% and 25.24% respectively. Furthermore, the addition of MT reduced the contents of ROS and MDA in the algal cells and increased the activities of three antioxidant enzymes, and alleviated algal cell oxidative damage caused by nitrogen deficiency. The exogenous MT could further promote the accumulation of lipid in microalgae under nitrogen deficiency condition by regulating the ROS level and the antioxidant enzymes activities, which provided a novel strategy for the synthesis of lipid in microalgae.
