      采用间接免疫法结合显色反应快速检测食用油中苯并(a)芘,在抗体与磁珠上苯并(a)芘抗原结合的基础上,通过辣根过氧化酶连接苯并(a)芘二抗进行间接免疫反应。结果表明:优化的免疫磁珠反应条件为苯并(a)芘抗体用量4 μg,辣根过氧化酶用量100 μg,免疫反应温育时间60 min;方法的线性范围为0.5 pg/mL~500 ng/mL,检出限为0.12 μg/kg,定量限为0.5 μg/kg,加标回收率为89.3%~100.1%。方法具有良好的特异性和稳定性,符合食品污染物现场快速检测的基本要求。
英文摘要:
      Indirect immuneassay combined with color reaction was used for the rapid detection of benzo(a)pyrene in edible oil. Based on the binding of antibody to benzo(a)pyrene antigen on magnetic beads, the indirect immune response was carried out by ligating benzo(a)pyrene antibody with horseradish peroxidase. The results showed that the optimal reaction conditions of immune magnetic beads were obtained as follows: dosage of benzo(a)pyrene 4 μg, dosage of horseradish peroxidase 100 μg, and incubation time of immune reaction 60 min. The linear range,limit of detection, limit of quantitation and standard addition recovery of the method were 0.5 pg/mL-500 ng/mL, 0.12 μg/kg, 0.5 μg/kg and 89.3%-100.1%. The method had good specificity and stability, and met the basic requirements of rapid detection of food contaminants in the field.
