      为了研究高山被孢霉突变株12-2-2产花生四烯酸(ARA)的培养条件,选取温度、pH、摇床转速和培养时间为4个影响因素,以生物量、油脂产量和ARA产量为综合评价指标,进行单因素实验和正交实验。结果表明,影响高山被孢霉突变株12-2-2产ARA的因素主次顺序为:温度﹥培养时间﹥摇床转速﹥pH;最优培养条件为:温度26℃,培养时间7 d,摇床转速160 r/min,pH 8。在最优培养条件下,高山被孢霉突变株12-2-2的生物量为32.31 g/L,油脂产量为12.98 g/L,ARA产量为5.12 g/L。
英文摘要:
      With temperature, pH, shaking speed and culture time as four influencing factors, and biomass and productivities of oil and arachidonic acid (ARA) as comprehensive evaluation indexes, the culture conditions of Mortierella alpina mutant 12-2-2 to produce ARA were studied by single factor experiment and orthogonal experiment. The results showed that temperature had the greatest influence on the ARA production by Mortierella alpina mutant 12-2-2, followed by culture time, shaking speed and pH; the optimal culture conditions were obtained as follows: temperature 26℃, culture time 7 d, shaking speed 160 r/min and pH 8. Under these conditions,the biomass and productivities of oil and ARA were 32.31 g/L, 12.98 g/L and 5.12 g/L, respectively.
