      以羧甲基壳聚糖为水溶性包覆材料对乙醇注入法制备的薏苡仁油常规脂质体进行包覆,在单因素试验的基础上,以包覆率为指标,采用正交试验对薏苡仁油包覆脂质体的配方进行优化。确定最佳配方为:羧甲基壳聚糖(CMCT)与磷脂(PC)质量比5∶4,芯材添加量15%,磷酸盐缓冲液pH 6.5,乳化剂添加量0.04%(吐温80与聚乙二醇400质量比1∶4)。在此条件下制备的薏苡仁油包覆脂质体的包封率为86.78%,包覆率为99.18%,平均粒径为145.6 nm,Zeta电位为-30.4 mV,透射电镜观察可见明显的CMCT层包覆于常规脂质体表面,且包覆脂质体的稳定性明显高于常规脂质体。

      Conventional liposomes of coix seed oil prepared by ethanol injection was coated by carboxymethyl chitosan (CMCT). The preparation of CMCT-coated liposomes containing coix seed oil was optimized by orthogonal experiment with coating efficiency as index on the basis of single factor experiment. The results indicated that the optimal preparation conditions were as follows: mass ratio of CMCT to phospholipid (PC) 5∶4, core material dosage 15%, phosphate buffer pH 6.5, emusifier dosage 0.04%(mass ratio of tween 80 to polyethylene glycol 400 1∶4). The CMCT-coated liposomes containing coix seed oil obtained under the optimal conditions were characterized with high embedding efficiency (86.78%), heterogeneous size distribution (145.6 nm), eligible Zeta potential (-30.4 mV) and excellent coating efficiency (99.18%). In addition, transmission electron microscopy showed that these coated liposomes were spherical particles with the existence of CMCT layers surrounding on the surface of conventional liposomes. The CMCT layers could enhance the stability of conventional liposomes greatly.
