The crude tea saponin was separated and purified by macroporous resin. Using the yield of tea saponin as indicator, the elution conditions were optimized by response surface methodology. S-8 macroporous resin was screened out by studying the adsorption and desorption properties of macroporous resins D-101, AB-8, S-8, X-5 and HPD-100. The optimal adsorption conditions were obtained as follows: dosage of S-8 macroporous resin 100 g, mass concentration of tea sapoin solution 0.703 mg/mL, loading volume 110 mL and flow rate of sample 3.03 mL/min, volume fraction of ethanol (eluent)90%, flow rate 3.03 mL/min, dosage of ethanol 100 mL. After 47.38% of tea saponin was separated and purified under the optimal conditions, the yield, recovery rate and content of tea saponin were 81.74%, 78.29% and 85.36%,respectively. |
