With temperature, pH, shaking speed and culture time as four influencing factors, and biomass and productivities of oil and arachidonic acid (ARA) as comprehensive evaluation indexes, the culture conditions of Mortierella alpina mutant 12-2-2 to produce ARA were studied by single factor experiment and orthogonal experiment. The results showed that temperature had the greatest influence on the ARA production by Mortierella alpina mutant 12-2-2, followed by culture time, shaking speed and pH; the optimal culture conditions were obtained as follows: temperature 26℃, culture time 7 d, shaking speed 160 r/min and pH 8. Under these conditions,the biomass and productivities of oil and ARA were 32.31 g/L, 12.98 g/L and 5.12 g/L, respectively. |
