      建立了棉籽粕中游离棉酚(FG)的高效液相色谱法(HPLC)测定。结果表明,HPLC测定FG的最佳色谱条件为:流动相V(甲醇+0.2%磷酸)∶V(V(乙腈)∶V(水)=1∶3)=85∶15(分别使用2个进样器),柱压16.2 MPa,柱温25℃,流速1.0 mL/min,进样量20 μL,Waters XTerra MS C18不锈钢色谱柱(250 mm×4.6 mm,5 μm),UV检测波长235.5 nm。在最佳条件下,能更好地检测棉籽粕中的游离棉酚,棉酚标准溶液的质量浓度与其峰面积呈正相关,相关系数为0.999 9,在0.15~5 μg/mL之间呈良好线性关系,平均回收率在95.67%~98.99%之间,相对标准偏差在1.28%~2.03%之间。

      The determination method of free gossypol (FG) in cottonseed meal was established by high performance liquid chromatography (HPLC). The results showed that the optimal HPLC conditions were obtained as follows:mobile phase V(methanol +0.2% H3PO4)∶V(V(acetonitrile)∶V(water )=1∶3)=85∶15 (respective using two injectors),column pressure 16.2 MPa,column temperature 25℃,flow rate 1.0 mL/min,injection volume 20 μL,Waters XTerra MS C18 stainless steel chromatographic column (250 mm×4.6 mm,5 μm),UV detection wavelength 235.5 nm. The FG in cottonseed meal could be well detected under the optimal conditions. A good linear relationship between peak area and mass concentration of standard gossypol solution was observed in the range of 0.15-5 μg/mL(R2=0.999 9). The mean recovery was 95.67%-98.99% with relative standard deviation ranging from 1.28% to 2.03%.
