Determination of free gossypol in cottonseed meal by HPLC |

Abstract: |
The determination method of free gossypol (FG) in cottonseed meal was established by high performance liquid chromatography (HPLC). The results showed that the optimal HPLC conditions were obtained as follows:mobile phase V(methanol +0.2% H3PO4)∶V(V(acetonitrile)∶V(water )=1∶3)=85∶15 (respective using two injectors),column pressure 16.2 MPa,column temperature 25℃,flow rate 1.0 mL/min,injection volume 20 μL,Waters XTerra MS C18 stainless steel chromatographic column (250 mm×4.6 mm,5 μm),UV detection wavelength 235.5 nm. The FG in cottonseed meal could be well detected under the optimal conditions. A good linear relationship between peak area and mass concentration of standard gossypol solution was observed in the range of 0.15-5 μg/mL(R2=0.999 9). The mean recovery was 95.67%-98.99% with relative standard deviation ranging from 1.28% to 2.03%. |
