Effects of different oils on the spatial learning and memory of rats and the relationship with ASICs subtype receptor

Abstract:
      The influences of DHA oil,walnut oil,safflower oil,perilla oil and linoleic acid on the spatial learning and memory of weaned rats and its relationship with the expression of ASICs(acid sensing ion channels)gene were studied and discussed. A total of 48 weaned SD rats were divided randomly into 6 groups: negative control group(basal feed),DHA oil group(0.902 mL/(kg·d)),walnut oil group (2.445 mL/(kg·d)),safflower oil group(1.792 mL/(kg·d)),perilla oil group(2.445 mL/(kg·d)) and linoleic acid group (1.464 mL/(kg·d)).The spatial learning and memory ability was detected by Morris water maze test after the rats were administrated for 8 weeks,and the mRNA expression of ASICs in the hippocampus of rats was detected by RT-PCR. The results showed that compared with the negative control group, the escape latencies of groups of DHA oil,walnut oil, safflower oil,perilla oil and linoleic acid significantly shortened (P<0.05) and across platform times markedly increased (P< 0.05); compared with the negative control group,DHA oil,walnut oil,safflower oil,perilla oil and linoleic acid could not only significantly improve the expression of ASIC4 mRNA (P<0.05) in the brain hippocampus of rats, but also remarkably reduce the expressions of ASIC1a mRNA and ASIC1b mRNA (P<0.05 or P<0.01). DHA oil could evidently promote the expression of ASIC3 mRNA (P< 0.05). Walnut oil could markedly increase the expression of ASIC2b mRNA (P<0.05). Safflower oil could extremely enhance the expression of ASIC2b mRNA (P<0.01). Perilla oil could significantly improve the expressions of ASIC2a mRNA(P< 0.01) and ASIC2b mRNA (P<0.05). Linoleic acid could dramatically increase the expression of ASIC2a mRNA (P< 0.01). Therefore, DHA oil,walnut oil,safflower oil,perilla oil and linoleic acid could promote the ability of spatial learning and memory of rats and the promotion process of the ability of spatial learning and memory of rats by different oils was associated with the expression of ASICs subtype receptor.
