Sesamin was extracted by ultrasound-assisted 90% ethanol solution. On the basis of single factor experiment, the parameters of the ultrasound-assisted extraction of sesamin were optimized by orthogonal experiment. The reducing activity and the antibacterial activity of sesamin were determined by Prussian blue method and filter paper-inhibition zone method, respectively. The results showed that the optimal ultrasound-assisted extraction conditions were obtained as follows:ultrasonic power 300 W, ratio of liquid to solid 25∶1, ultrasonic temperature 55℃ and ultrasonic time 25 min. Under the optimal conditions, the extraction rate of sesamin was (3.942±0.085) mg/g. Sesamin had certain reducing activity, much weaker than VC of the same mass concentration. When the mass concentration of sesamin was higher than 40 mg/mL, sesamin showed different degrees of inhibitory action on Bacillus cereus,Staphylococcus aureus,Bacillus subtilis and Escherichia coli, and the inhibitory action of sesamin with mass concentration higher than 60 mg/mL on Bacillus cereus was the strongest, followed by Staphylococcus aureus,Bacillus subtilis and Escherichia coli. When the mass concentration of sesamin was less than 100 mg/mL, no obvious inhibitory action of sesamin on Aspergillus flavus was observed. |
