The injury of α-linolenic acid on HepG2 cell was investigated by MTT colorimetric method, inverted phase contrast microscope was used to observe morphology of HepG2 cell and the reactive oxygen species(ROS) level, superoxide dismutase(SOD) activity and malonadialdehyde(MDA) content were determined to discuss the effect of α-linolenic acid on the oxidative stress of HepG2 cell. The results showed that the proliferation of HepG2 cell was inhibited by α-linolenic acid in a dosage-dependent(50-250 μmol/L) and time-dependent manner, which indicated that α-linolenic acid could induce the injury of HepG2 cell. The morphology of HepG2 cell changed after incubated with 200 μmol/L α-linolenic acid for 24 h and showed typical apoptotic morphology changes in high concentration. After treatment with α-linolenic acid,the contents of ROS and MDA in HepG2 cell increased,while the activity of SOD decreased, indicating that α-linolenic acid affected the oxidative stress of HepG2 cell to induce the injury of HepG2 cell. |
