      采用复合磷脂酶在水相体系中酶解大豆卵磷脂制备甘油磷脂酰胆碱(GPC),磷脂酶A1和磷脂酶A2复合酶解作用,不仅有效地缩短了酶解时间,更能提高GPC的得率。通过响应面优化试验确定最佳酶解条件为:底物质量浓度61 g/L,酶解温度45.2℃,磷脂酶A1添加量16.86 U/mL,磷脂酶A2添加量21.98 U/mL,CaCl2添加量5.18 g/L。在最佳酶解条件下酶解2 h,GPC得率平均值为96.25%。

      Glycerylphosphorylcholine (GPC)was prepared by the enzymatic hydrolysis of soybean lecithin in aqueous media with complex phospholipase as catalyst, and the complex enzymatic hydrolysis by phospholipase A1 and phospholipase A2 could not only effectively shorten the enzymolysis time, but also improve the yield of GPC. The optimal enzymatic hydrolysis conditions were determined by response surface methodology as follows:mass concentration of substrate 61 g/L, enzymolysis temperature 45.2℃, dosage of phospholipase A1 16.86 U/mL, dosage of phospholipase A2 21.98 U/mL, dosage of CaCl2 5.18 g/L. Under the optimal conditions, the average yield of GPC was 96.25%after enzymatic hydrolysis for 2 h.
