Response surface methodology was applied to optimize the microwave-ultrasound-assisted extraction conditions of flaxseed gum so as to improve the extraction rate and yield of flaxseed gum. Meanwhile, the composition of flaxseed gum was analysed by FTIR. Using ratio of liquid to material, extraction temperature and extraction time as influence factors, and yield of flaxseed gum as response value, the quadratic polynomial regression equation was established by the Box-Behnken design on the basis of single factor experiment. The results showed that the optimal extraction conditions of flaxseed gum were obtained by response surface methodology as follows: extraction temperature 85 ℃, ratio of liquid to material 17.3∶1 and total extraction time 65 min. Under these conditions, the yield of flaxseed gum was 5.14%, closed to the theoretical value (5.33%). The FTIR result indicated that flaxseed gum was mainly composed of polysaccharides and protein. |
