      采用单因素试验和正交试验对杜仲籽蛋白的提取及纯化工艺条件进行优化,并分析了杜仲籽蛋白的氨基酸组成及含量。结果表明:杜仲籽蛋白最佳提取条件为浸提时间50 min、浸提温度45℃、料液比1∶20、浸提次数2次、搅拌速度40 r/min、亚硫酸氢钠添加量0.25%、酸沉pH 3.9,杜仲籽蛋白最佳纯化条件为洗涤次数3次、洗涤液乙醇体积分数85%、料液比1∶6、洗涤温度-10℃、洗涤时间30 min;在最佳提取和纯化条件下,杜仲籽蛋白提取率为85.6%,纯化得率为97.6%。经测定杜仲籽蛋白含有18种常见氨基酸,其中人体必需氨基酸种类齐全,且含量较高。

      The extraction and purification conditions of Eucommia ulmoides seed protein were optimized by single factor experiment and orthogonal experiment,and the amino acid composition and content ofEucommia ulmoides seed protein were analyzed. The results showed that the optimal extraction conditions were obtained as follows: extraction time 50 min, extraction temperature 45℃, ratio of solid to liquid 1∶20,extraction times twice, stirring speed 40 r/min, dosage of sodium bisulfite 0.25%, acid precipitation pH 3.9; and the optimal purification conditions were obtained as follows: washing times 3 times, volume fraction of ethanol 85%, ratio of solid to liquid 1∶6, washing temperature -10℃, washing time 30 min;under the optimal conditions, the extraction rate and purification yield of Eucommia ulmoides seed protein were 85.6% and 97.6%, respectively. 18 amino acids were detected fromEucommia ulmoides seed protein,in which the variety of essential amino acid was complete and the content was high.
