Abstract: |
The nutrients promoting the production of γ-linolenic acid by Mortierella isabellina were found by studying on metabolic regulation of production process of γ-linolenic acid. The medium nutrients were screened by Plackett-Burman experiment.The results showed that the key factors influencing oil yield were glucose,KNO3,sodium acetate and sodium citrate.The four key factors were optimized by response surface methodology with yield of γ-linolenic acid as response value.Combining Plackett-Burman experiment with response surface methodology,the optimal medium formula were obtained as follows:glucose 100.90 g/L,KNO3 1.57 g/L,sodium acetate 3.42 g/L,sodium citrate 4.56 g/L,yeast extract 3.75 g/L,KH2PO42.25 g/L,MgSO4·7H2O 0.60 g/L,CaCl20.30 g/L,FeSO40.15 g/L,ZnSO40.20 g/L.Under the optimal conditions,the yield of γ-linolenic acid reached 1 525.20 mg/L,increasing by 1.1 times than the initial yield (725.90 mg/L) . |
