Schizochytrium’s cell structure is tight, so wall-breaking is the key process of DHA high strength fermentation.With Schizochytrium as experimental bacteria, the wall-breaking methods, including water-washing,membrane filtration, high pressure homogeneous, enzymolysis and high pressure homogeneous composite membrane filtration were compared, and the morphological characteristics of cells before and after wall-breaking by each method were observed, in the same time the cell wall-breaking rate, oil content of fermentation liquid, and productivity and content of DHA were determined. The results showed that the effect of high pressure homogeneous composite membrane filtration method was the best with wall-breaking rate 100%, also the oil content of fermentation liquid, and productivity and content of DHA of that method were the highest, reaching 16.98,7.08 g/L and 41.71%,respectively. So high pressure homogeneous composite membrane filtration method was the best way to break the wall from the aspect of continuity and efficiency of production. |
