An oleaginous yeast was screened from sea mud and algae attached rock by Sudan Black B staining method and it was identified as Meyerozyma guilliermondii by physicochemical and biochemical characteristics combined with molecular biological methods. The oil was extracted by acid-heating method. Based on single factor experiment, the extraction conditions were optimized by orthogonal experiment. The optimal extraction conditions were obtained as follows: 0.5 g of dried cells, 4 mol/L HCl 10 mL, acid-heating time 10 min, organic solvent(volume ratio of chloroform to methanol 2∶?1) 25 mL, 0.15% NaCl 10 mL. Under these conditions, the oil yield was 40.12%. |
