      验证了磷酸香草醛反应在不同氮(0.2、2、20、40 mg/L)、磷(0.1、1、10、20 mg/L)质量浓度培养条件下测定四尾栅藻总脂含量的可靠性。结果表明:不同氮、磷质量浓度条件下栅藻总脂含量和磷酸香草醛测定吸光度之间均呈显著的线性回归关系(不同氮质量浓度条件:R=0.997,P=0.003;不同磷质量浓度条件:R=0.994,P=0.006),在不同氮、磷质量浓度培养条件下该方法的测定结果准确可靠,可以用于栅藻的总脂含量测定。磷酸香草醛反应的显色强度可能与脂肪酸的饱和度有关,为保障该方法测定总脂含量的准确性,用该方法估算总脂含量须在氮、磷质量浓度变化范围内进行预先验证。

      The reliability of measuring total fat content of Scenedesmus obliquus using the phosphoric acid-vanillin reaction under different nitrogen (0.2, 2, 20, 40 mg/L) and phosphorus (0.1, 1, 10, 20 mg/L) mass concentration cultural conditions was studied. The results showed that there were significantly linear regression relationships between total fat content and absorbance value of phosphoric acid-vanillin determination under different nitrogen and phosphorus mass concentrations (different nitrogen mass concentration conditions: R=0.997, P=0003; different phosphorus mass concentration conditions: R=0.994, P=0.006). This method was accurate and reliable under different phosphorus and nitrogen mass concentrations. It could be used to determine the total fat content of Scenedesmus. The color intensity of phosphoric acid-vanillin reaction was associated with the saturation degree of fatty acids. Therefore, the range of nitrogen and phosphorus mass concentration should be verified to ensure the accuracy before using this method.
