Abstract: |
The reliability of measuring total fat content of Scenedesmus obliquus using the phosphoric acid-vanillin reaction under different nitrogen (0.2, 2, 20, 40 mg/L) and phosphorus (0.1, 1, 10, 20 mg/L) mass concentration cultural conditions was studied. The results showed that there were significantly linear regression relationships between total fat content and absorbance value of phosphoric acid-vanillin determination under different nitrogen and phosphorus mass concentrations (different nitrogen mass concentration conditions: R=0.997, P=0003; different phosphorus mass concentration conditions: R=0.994, P=0.006). This method was accurate and reliable under different phosphorus and nitrogen mass concentrations. It could be used to determine the total fat content of Scenedesmus. The color intensity of phosphoric acid-vanillin reaction was associated with the saturation degree of fatty acids. Therefore, the range of nitrogen and phosphorus mass concentration should be verified to ensure the accuracy before using this method. |
