Abstract: |
A HPLC method was developed for the separation of 1-laurin-3-palmitin diacylglycerol and 1-laurin-2-palmitin diacylglycerol. The optimal chromatographic analysis conditions were obtained as follows: evaporative light scattering detection (ELSD) as detector (drift tube temperature 90℃ and N2 flow rate 2.2 L/min), mobile phase of acetonitrile-isopropanol (volume ratio 95∶5), flow rate 1.0 mL/min, column temperature 35℃. Under the optimal chromatographic analysis conditions, the standard curve equation of 1-laurin-3-palmitin diacylglycerol (mass concentration range 0.05-5.00mg/mL) was Y = 1.832X + 4.564 with a good linear regression coefficient (R2=0.998 8). The method showed good reproducibility, precision and accuracy, which was beneficial for the qualitative and quantitative analysis of diacylglycerol positional isomers in structural lipids. |
