The contents of free and conjugated α-linolenic acid, linoleic acid and oleic acid in Paeonia suffruticosa Andr. seed oil were determined by HPLC. The detection conditions were as follows: Agilent C18 column (3.0 mm×50 mm, 2.7 μm), mobile phase acetonitrile-0.1% phosphoric acid solution (volume ratio 85∶15), flow rate 1 mL/min, detection wavelength 203 nm, column temperature 35℃ and injection volume 20 μL. The results showed that the method had good precision, repeatability and stability. Good linear relationships of α-linolenic acid, linoleic acid and oleic acid were achieved in the mass concentration ranges of 41.4-207 μg/mL, 20.2-101 μg/mL and 14.9-74.6 μg/mL, respectively. The average contents of free α-linolenic acid, linoleic acid and oleic acid were 4.47,1.89 mg/mL and 0.92 mg/mL respectively, and the average contents of conjugated α-linolenic acid, linoleic acid and oleic acid were 437,237 mg/mL and 156 mg/mL respectively in Paeonia suffruticosa Andr.seed oil. The methods had the advantages of high separation efficiency, good selectivity and mild conditions. |
