Abstract: |
An oleaginous yeast (strain 114) utilizing xylose was screened and isolated from nature environment by the phosphoric acid-vanillin reaction. This strain was identified based on physiological and biochemical characterization and 26S rDNA sequence analysis. The fermentation conditions of strain 114 were optimized, and the effects of culture time, inoculation amount, fermentation temperature and media amount on oil production capacity of the strain were investigated. The fatty acid composition of the obtained oil was analyzed by GC-MS. The results showed that the strain was identified as Rhodotorula mucilaginous, and it grew well in enzymolysis liquid of Rhodotorula mucilaginous spent grain. The optimal fermentation conditions were obtained as follows: culture time 5 d, inoculation amount 12%, fermentation temperature 32℃ and media amount 30 mL/250 mL. Under these conditions, the biomass, oil yield and oil content were 16.92, 4.44 g/L and 26.22% respectively. The contents of unsaturated fatty acid and oleic acid were 76.923% and 59.087% respectively, which were similar to vegetable oil. |
