Waste cooking oil was hydrolyzed to prepare fatty acids with lipase from different sources as catalyst.The dosage of lipase was studied by single factor experiment, and on this basis, the dosage of lipase, ratio of oil to water, reaction temperature and reaction time were optimized by orthogonal experiment. The results showed that waste cooking oil could be hydrolyzed successfully both by Porcine pancreas lipase L3621 and Candida rugosa lipase LS20. The optimal conditions were obtained as follows: dosage of lipase 700 U/g, reaction temperature 45?℃, reaction time 36 h and ratio of oil to water 1∶?1.2 for L3621, and under these conditions, the enzymatic hydrolysis rate reached 94.30%; dosage of lipase 600 U/g, reaction temperature 40?℃, reaction time 36 h and ratio of oil to water 1∶?1 for LS20, and under these conditions, the enzymatic hydrolysis rate reached 96.84%. |
