Abstract: |
Sporobolomyces reseus As.2.618 was screened by two compound mutagenesis methods UV combined with NTG and UV combined with DES to obtain high oil yield strain. UV combined with NTG added by maleic hydrazide was applied to obtain UV-NTG-2 strain,its biomass, oil content and oil yield reached 39.01 g/L, 19.83% and 7.74 g/L respectively, increasing by 42.42%, 52.30% and 11681% compared with the original strain. UV-DES-2 strain was mutated by UV combined with DES and screened by cerulenin.Its biomass, oil content and oil yield reached 38.96 g/L, 20.80% and 8.10 g/L respectively, increasing by 42.24%, 59.75% and 126.89% compared with the original strain. In addition, the relative contents of C18∶1 in the two mutant strains oils increased by 11.88 percentage points and 6.69 percentage points compared with the original strain, respectively. Furthermore, both of their relative contents of C20∶1 decreased, which indicated that the oil fluidity of the mutant strains improved. The mutant strains had stable genectic characters after five serial passages. |
