Abstract: |
The preparation process of lycopene-loaded nanostructured lipid (lyco-NLC) was studied. With product average particle size, polydispersity index(PDI) and encapsulation efficiency as indexes,the influences of material and operation factors on lyco-NLC were studied using single factor experiment and orthogonal experiment to obtain the optimal preparation process. The morphology of lyco-NLC was observed, and its storage stability was studied. The results showed that the optimal preparation process conditions were obtained as follows: total lipid mass concentration 7.5 g/100 mL, mass ratio of solid lipid to liquid lipid 3.5∶?1, surfactant Tween 80 mass concentration 2 g/100 mL, homogenization pressure 50 MPa, homogenization times 3. Under these conditions, the average particle size of lyco-NLC was 230 nm with PDI 0.23, the encapsulation efficiency was 91.95% and the lycopene loading amount in the product was up to 4%. The product possessed good water dispersibility. Particles of the lyco-NLC were found to be spherical and uniform in shape under transmission electron microscopy (TEM).The 30 d storage experiment results showed that lyco-NLC had good physical stability (average particle size <270 nm, PDI <0.3). |
