Abstract:
      In order to improve the tropical rich-in-oil microalgae Desmodesmus sp. WC08 oil yield, the response surface methodology was adopted for autotrophic study on optimization of culture conditions. By single factor experiment, nine factors optimal concentrations which could promote oil yield of Desmodesmus sp. WC08 were obtained. Based on Plackett-Burman experiment, four significant factors were screened owing to Desmodesmus sp. WC08 oil yield. The optimal conditions were determined through the Box-Behnken experiment and response surface methodology as follows: nitrogen concentration 10 mmol/L, salinity 1.6%, initial OD680 0.33, initial pH 6.5. Under the optimal conditions for cultivating Desmodesmus sp. WC08, the oil yield was 141.47 mg/(L·d), increased by 147.33% than that without optimization.
