Abstract: |
The culture medium and culture conditions of the mutant strain Mortierella ramanniana HLY0902 were optimized by orthogonal experiment and single factor experiment in order to increase the production of gamma-linolenic acid(GLA). The results showed that the yield of GLA was up to 1.05 g/L in the medium containing glucose 100 g/L, yeast extract powder 10 g/L, KH2PO4 4 g/L, NaNO3 1 g/L, MgSO4 ·7H2O 0.5 g/L and increased by 43.8% compared with that before optimization. The optimal culture conditions were identified as follows: inoculation amount 10%, loading amount 20%, batch time 168 h. Furthermore, the optimal conditions for mycelium growth and oil accumulation were pH 5.5 and 22℃, and the optimal conditions for GLA accumulation were pH 7.5 and 20?℃. The results showed that the capacity of GLA production by mutant strain Mortierella ramanniana HLY0902 improved through the optimization. |
