Abstract: |
By optimizing the extraction conditions of soybean water-soluble protein and using Dumas combustion method, an determination method of soybean water-soluble protein content was established, and the optimal experimental conditions of the Dumas combustion instrument were discussed. The results showed that the optimal extraction and drying conditions were obtained as follows: solid-liquid ratio 1∶?30 at constant temperature 20?℃, extraction times twice, extraction time 40 min, extract volume 3-4 mL when the extract evaporated and dried, and electric heating plate temperature 150 ?℃. The optimal experimental conditions of the Dumas combustion instrument were obtained as follows: oxygen flow rate 300 mL/min, and oxygen factor 1.4 mL/mg. Under the optimal conditions, the test precision of this method met the technical requirements of laboratory variation coefficient (CV) during the validation of the test method in GB/T 27404—2008, and there was no significant difference from the current Kjeldahl test method (P=0.89). The method belonged to the environment-friendly chemical analysis method and could meet the pollution-free, rapid and accurate detection of water-soluble protein in soybean. |
