Cloning and expression analysis of 3-ketoacyl-CoA synthase gene from Malania oleifera |

Abstract: |
The Malania oleifera was used as material to clone the full length cDNA of the 3-ketoacyl-CoA synthase (KCS) gene based on the transcriptome of Malania oleifera with the method of RT-PCR technology, and named MoKCS1(GenBank ID: MK210592). The sequence analysis of MoKCS1 gene showed that the full-length cDNA of MoKCS1 gene was 1 539 bp, encoded 512 amino acids, and belonged to KCS family. The proteinic sequence alignment revealed that there was 80.66% of similarity between Malania oleifera and Durio zibethinus, and MoKCS1 had three functionally conserved domains that were unique in the KCS family. The phylogenetic analysis with known ultralong-chain KCS proteins revealed that no protein was found to be closely related to MoKCS1. The fluorescent quantitative RT-PCR certified that the expression of MoKCS1 gene in fruit of the flower within 3 months was the highest. |
