Traditional solid immobilized lipase has problems such as fragile carrier, high preparation cost etc., which restricts its industrial application. Three liquid phase system developed in the previous period was used as a liquid immobilized carrier for lipase.The effects of different kinds of phase-forming substances, the proportion of phase-forming substances and different kinds of lipases on the recovery of immobilized liquid enzyme were studied. And the stability of the lipase immobilized on the liquid immobilized carrier was investigated. The results showed that the middle phase of n-hexane/PEG400/Na2SO4 system was a better liquid immobilized lipase carrier. The optimal conditions were obtained as follows: PEG400 ratio 20%, and Na2SO4 ratio 14%. Under these conditions, the recovery of lipase AY30 could be over 98%, and the recoveries of other types of lipases were all more than 90%. The liquid immobilized carrier could effectively maintain the catalytic activity of the lipase at 30?℃, and the protective effect was obviously superior to the traditional water-organic solvent system at different temperatures. At the same time, the preservation stability of the lipase in the liquid immobilized carrier was also better than that in the water environment. After two weeks of low temperature preservation, it could still retain about 78.4% of the enzyme activity, which showed good preservation stability. |
