In order to explore the disruption effect of physical ball milling method with mature industrial conditions on R. toruloides cell, with oil yield as the evaluation index, the effects of four critical factors including milling media, milling time, ball mill speed and dosage of media were studied. On the basis of single factor experiment, the optimal cell disruption process conditions were obtained by response surface methodology as follows: zirconium silicate bead used as milling media, milling time 2.5 h, ball mill speed 360 r/min and dosage of media (based on yeast liquor volume ) 41%. Under these conditions, the oil yield was 57.32%. |
