Indirect immuneassay combined with color reaction was used for the rapid detection of benzo(a)pyrene in edible oil. Based on the binding of antibody to benzo(a)pyrene antigen on magnetic beads, the indirect immune response was carried out by ligating benzo(a)pyrene antibody with horseradish peroxidase. The results showed that the optimal reaction conditions of immune magnetic beads were obtained as follows: dosage of benzo(a)pyrene 4 μg, dosage of horseradish peroxidase 100 μg, and incubation time of immune reaction 60 min. The linear range,limit of detection, limit of quantitation and standard addition recovery of the method were 0.5 pg/mL-500 ng/mL, 0.12 μg/kg, 0.5 μg/kg and 89.3%-100.1%. The method had good specificity and stability, and met the basic requirements of rapid detection of food contaminants in the field. |
