Separation of 37 fatty acid components by two chromatographic columns |

Abstract: |
Thirty-seven fatty acid standard samples and oil samples were separated by CD-2560 chromatographic column and DB-WAX chromatographic column respectively. The effects of flow rate,split ratio and temperature programming on the separation effect of the chromatographic column were explored. In addition,the peak order and the name of each group in the 37 fatty acid mixtures separated by DB- WAX were determined by the known CD-2560 chromatographic column for the peak order,standard sample mass concentration and oil sample comparison. The results showed that the optimal separation conditions of these two kinds of chromatographic columns were obtained as follows: mass concentration of mixed standard 1 mg/mL, column flow rate 0.55 mL/min, split ratio 70∶ 1, temperature programmed from 100 ℃ (retaining for 5 min) to 230 ℃ at 3 ℃/min rate (CD-2560 chromatographic column retaining for 30 min, DB-WAX chromatographic column retaining for 50 min).Using these two chromatographic columns to determine the fatty acid content in five oil samples, the separation results were almost identical, which meant that the CD-2560 chromatographic column and the DB-WAX chromatographic column could be applied to determine 37 fatty acids in the vegetable oil. |
