Abstract: |
The protective effect of Gardenia oil on acetaminophen(AP)-induced acute liver injury in mice and mechanism were investigated. The mice were randomly divided into control group, model group, biphenyl dimethylester group, and low, middle and high dose groups of Gardenia oil, and administrated once a day for 7 d,and 1 h after the last administration, the mice were intraperitoneally injected with AP of 450 mg/kg in each group except for the control group.The contents of ALT, AST and ALB in the blood serum, the activity of GSH and SOD, and the level of MDA in the liver organs of mice were determined, and pathological morphological changes in liver cells were monitored by HE staining. The results showed that compared with model group, each dose group of Gardenia oil could reduce the contents of ALT and AST, improve the content of ALB and the activities of GSH and SOD, and reduce the production of MDA in liver tissue, pathological damage in liver such as liver tissue degeneration and necrosis were significantly improved, especially in the high dose group of Gardenia oil. The results suggested that Gardenia oil had markedly protective effect on AP-induced acute liver injury in mice, and the mechanism may be related to its ability of improving the activity of antioxidant enzyme system, inhibiting lipid peroxidation and maintaining normal morphology of liver cell membrane. |
