      在无溶剂体系下,以棕榈硬脂和油酸为原料,sn-1,3位特异性脂肪酶为催化剂,合成富含OPO的人乳替代脂。采用单因素实验对反应条件进行优化,并采用高效液相色谱与气相色谱对反应产物的甘油三酯和sn-2位脂肪酸组成进行分析。结果表明:酶法催化酸解反应的优化条件为反应时间4 h、底物摩尔比1∶ 10、反应温度60 ℃、脂肪酶NS40086用量12%;在优化条件下,所得产品的OPO含量可达72.53%,sn-2位棕榈酸含量为87.03%。

      :Using palm stearin and oleic acid as raw materials, human milk fat substitute rich in OPO was prepared by enzymatic acidolysis in solvent-free system with sn-1, 3-position specific lipase as catalyst. The reaction conditions were optimized by single factor experiment and the compositions of triglyceride and fatty acid in sn-2 position of the substitute were analyzed by high performance liquid chromatography and gas chromatography. The results showed that the optimal conditions of enzymatic acidolysis reaction were obtained as follows: reaction time 4 h, molar ratio of substrate 1∶ 10, reaction temperature 60 ℃ and dosage of lipase NS40086 12%. Under these conditions, the contents of OPO and palmitic acid in sn-2 position reached 72.53% and 87.03%, respectively.
