Abstract: |
Epidemiological study has reported that trans palmitoleic acid (9t16∶ 1),the representative trans fatty acid in ruminants,has the effect of reducing blood lipid. To further investigate the effect of 9t16∶ 1 on lipid metabolism in cells,different concentrations of 9t16∶ 1 were adopted to treat steatotic human hepatocytes LO2 in the experiment, and the indexes of the cell viability, intracellular fatty acid composition, lipid content changes and cell lipid oxidation were determined. The results showed that the content of saturated fatty acids decreased by 34.89%, the content of monounsaturated fatty acids increased by 16.44% and the content of polyunsaturated fatty acids increased by 30.50% after human hepatocytes being treated with 9t16∶ 1 of 100 μmol/L.9t16∶ 1 Partly transformed into 11t18∶ 1 and CLA in human hepatocytes, and the conversion rates were 17.50% and 16.42% respectively.9t16∶ 1 Significantly reduced the contents of triglyceride and cholesterol in steatotic human hepatocytes with the reduction rates of 61.76% and 37.80%, respectively.In addition, 9t16∶ 1 promoted intracellular lipid oxidation by significantly increasing the MDA level(increased by 45.36%) and decreasing SOD activity (decreased by 74.42%)in human hepatocytes.Therefore, 9t16∶ 1 could reduce lipid accumulation by promoting lipid oxidation in human hepatocytes. |
