Abstract: |
Transcriptome analysis for the original Schizochytrium limacinum SR21 strain and ARTP mutagenic strain NA2 with high yield of DHA was implemented with RNA-seq. The function of genes was annotated and metabolic pathways was classified to look for different expression genes and classify these genes into related metabolic pathways to reveal the mechanism of high yield of DHA in NA2. The results showed that 314 genes were up-regulated and three genes were down-regulated compared with the original strain. The up-regulated genes were mainly involved in amino acid metabolism and energy metabolism, which provided more acetyl-CoA and NADPH for the accumulation of polyunsaturated fatty acids. Validating the differently expressed genes with realtime fluorescence quantitative PCR, the result was in accordance with the transcriptome analysis. |
